匍枝筋骨草再生体系构建

Study on the Regeneration System of Ajugalobata

摘要:

为了提高匍枝筋骨草繁殖速度,在短期内获得大量试管苗,为种质资源的安全和有效保存提供技术指导。本研究以匍枝筋骨草为试验材料进行组培试验。用匍枝筋骨草叶片培养获得了组培苗,建立起了良好的扩繁体系。结果表明,采用匍枝筋骨草嫩叶为外植体,3% NaClO处理10 min可达到较好的消毒效果。诱导不定芽增值的最适培养基为MS+6-BA 1 mg/L,增殖率高;诱导愈伤的最适培养基为MS+6-BA 1 mg/L+2,4-D 0.4 mg/L,所得愈伤组织大而绿,长势良好;诱导生根的最适培养基是MS+IBA 0.4 mg/L,所得根系发达,植株健壮。本实验建立了完整的匍枝筋骨草植株再生体系,为匍枝筋骨草的大规模生产奠定了基础。

Abstract:

In order to increase the propagation rate for mass-produced Ajuga lobata and provide the technical guidance for the safety of germplasm resources and effectively preserve, the tissue culture and rapid propagation of Ajuga lobata was studied. The regeneration system of Ajuga lobata was well established by leaf blade. The results summarized as fellow: With 10 min NaClO 3% it can achieve good results by using leaves as explants. MS +6-BA 1 mg/L was the optimum multiplication culture medium. It can get high proliferation rate. MS+6-BA 1mg/L+2,4-D 0.4 mg/L was the optimum callus culture medium. The callus was large and green. MS+IBA 0.4 mg/L was the optimum culture medium taking roots. The roots were flourishing and the plant was strong. Thus the regeneration system of Ajuga lobata was established and it provided a foundation for Ajuga lobata mass production.
