马齿苋离体培养再生体系的建立

摘要:

以马齿苋幼嫩叶片和茎段为试验材料,对外植体灭菌、愈伤组织诱导、不定芽分化和试管苗生根的最佳条件进行研究,以期建立马齿苋再生技术体系,并为马齿苋的规模化繁殖和遗传改良等研究奠定基础。结果表明,最佳外植体灭菌方法为:用0.1%的升汞溶液浸泡8 min;最佳愈伤组织诱导培养基为MS+2,4-D 0.5 mg/L+6-BA 0.5 mg/L;最佳不定芽分化培养基为MS+6-BA 5.0 mg/L+NAA 0.4 mg/L;最佳生根培养基为1/2MS+IBA 2.0 mg/L。

Abstract:

To establish the regeneration technique system of Portulaca oleracea which lay the foundation for the research of large-scale propagation and genetic improvement, the optimal conditions for disinfecting of explants, inducing of callus, differentiating of adventitious buds and rooting of test-tube seedlings of Portulaca oleracea were studied by using leaf and stem bearing buds as explants. The results showed that the optimum disinfection method was to put explants in 0.1% HgCl2 for 8 min; MS+2,4-D 0.5 mg/L+6-BA 0.5 mg/L was the best medium to induce callus, MS+6-BA 5.0 mg/L+NAA 0.4 mg/L was the best sprout medium; The optimal culture medium was 1/2MS+IBA 2.0 mg/L for rooting.
