一种有效的菊花总RNA提取方法

An Effective Method for Extracting Total RNA of Chrysanthemum

摘要:

菊花组织或器官中含有大量的酚类、多糖、蛋白质等次生代谢物质,影响了菊花总RNA的提取质量。本研究用改良TRIZOL法提取菊花不同器官总RNA,并用凝胶电泳、紫外分光光度计对提取的总RNA质量和完整性进行检测,并对叶片总RNA进行反转录,通过RT-PCR扩增了菊花管家基因Actin的部分片断。结果表明,采用改良TRIZOL法提取的RNA条带清晰、完整性好、质量高,其中A260/A280比值在2.08~2.19。通过RT-PCR从叶片中扩增出了目的基因cDNA片断。该方法是一种快速、有效的提取菊花总RNA的方法;该方法适用于根、茎、叶、花等器官或组织总RNA的提取,可作为菊花不同组织和器官总RNA提取的通用方法。

Abstract:

Phenols, polysaccharide protein and other secondary metabolism substances existing in tissues or organs which influence total RNA extracting from Chrysanthemum. In this study, the total RNA from leaves. roots. shoots and flowers of Chrysanthemum were extracted by improved Trizol method and was detected by gel electrophorese and RT-PCR. The results showed that the total RNA were high purity and the strips of it was clear and complete. The A260/A280 detected by UV-Spectroscopic was 2.08-2.19. Partial gene of Actin was achieved by RT-PCR from leaves. The improved Trizol method was quick and efficiency for extracting total RNA from Chrysanthemum. It was a common method to isolate total RNA from different tissues and organs of Chrysanthemum.
