塔里木马鹿再生茸组织中原癌基因c-myc、c-fos的表达

Expression of Proto-oncogene c-fos, c-myc in different growth stages of Reborn Antlers of Tarim Deer’s Antler

摘要:

为进一步确定原癌基因c-myc、c-fos对马鹿茸生长的作用,实验收集231头2至4岁公鹿产茸量的生产信息,并采用3头成年马鹿生长期为30天、60天的头茬鲜茸和3头成年马鹿割后生长30天、60天的再生鲜茸,通过荧光定量PCR技术对不同组织基因的表达定量分析。结果表明,在鹿茸生长早期c-myc在再生茸茸皮的表达量显著低于头茬茸(P<0.01)。c-myc基因在割后再生的60天鹿茸茸皮和软骨组织表达下调,与头茬茸60天的相比,表达有显著差异(P<0.05)。间充质细胞层和成软骨层c-myc基因的表达量很低,且表达没有差异。从30天到60天,c-fos基因割后再生的马鹿茸茸皮中的表达下调,而在软骨组织中表达上调。与头茬60天茸相比,表达均有显著差异(P<0.05)。而在间充质细胞层和成软骨层没有差异。c-fos基因头茬与割后再生的60天马鹿茸的表达趋势与生长期为30天的相同。与头茬茸比较,再生茸成软骨层组织c-fos基因表达有上调趋势。结论:原癌基因c-myc、c-fos对茸皮的生长主要是促进其表皮干细胞的增殖分化;而对软骨组织c-myc基因高表达有阻碍茸软骨骨化,减缓骨化进程的作用,c-fos基因低表达有抑制生茸期骨膜内生骨的过程。

Abstract:

To study the effect of oncogene c-fos and c-myc on velvet antler growth, the antler yield of 231 Tarim wapitis were collected. And the first-cut antler and reborn fresh velvet antlers of 3 adult Tarim wapitis with 30 or 60-day-old were used to be experimental material. Quantitative expression in different zones of antler were determined by fluorescent quantitative PCR. The results demonstrated that the expression of c-myc in reborn dermal layer of antler early growth was lower than that in the first-cut antlers (P<0.01). The gene c-myc is down regulated in dermal layer and cartilage layer of the reborn antler with 60-day-old, and compared to 60d of natural growth antler, it were significant difference in both of them(P<0.05). The low expression of c-myc was observed in mesenchymal cells layer and precartilage cells layer in different growth period. Dropped expression of c-fos was observed in dermal layer of reborn antler while upward expression in cartilage layer from 30d to 60d and there was significant difference compared to natural growth antler(P<0.05).Little variation was found between the expressions of c-fos in mesenchymal cells layer and precartilage cells layer.Express of c-fos in reborn antlers with 60-day old was similar to that the first-cut antler with 30d, and c-fos is down regulated in cartilage layer of the reborn antler. The results suggested that c-myc and c-fos mainly promote the proliferation and differentiation of stem cells in dermal layer during the rapid growth of the antlers while in cartilage layer high expression of c-myc may inhibit cartilage osteoblast and low expression of c-fos inhibit process of Periosteal endogenous bone in antler’s growth period.
