油菜种子内生菌的去除及对萌发率的影响

Endophytic Bacteria Elimination in Rape Seed and Its Effect on Germination Rate

摘要:

为了研究内生菌对植物的生理作用,获得无内生菌干扰的植物种子,以油菜种子为材料,采用干热和湿热的方法去除种子固有内生菌。结果表明,50℃干热处理3天,60℃处理1~3天都能有效去除内生菌,去除率达99%以上,而种子的萌发率达86%,与对照组相比无显著变化;40℃湿热处理3天,50℃处理1~3天,内生菌的去除率99%以上,但湿热处理后种子的萌发率最高只有4%。因此采用干热处理是去除种子内生菌的有效方法,并为无菌苗的培养奠定基础。

Abstract:

In order to investigate the physiological effect the endophytic bacteria did on the plant, get the seeds without endophytic interference, in our research, we used the rape seeds as our material and we eliminated the natural endophytic bacteria of the rape seeds by dry heat treatment and moist heat treatment. Our results showed that, the treatment by dry beat sterilization at 50℃ for 3 days, or at 60℃ for 1-3 days could effectively eliminate the endophytic bacteria, the elimination rate could reach 99%, meanwhile, the seeds germination rate could reach 86%. But after moist heat treatment, the seeds germination rate was only 4%. We concluded that, dry hot treatment was the efficient method for eliminating the seed endophytic bacteria, and it would be the foundation for culturing the aseptic seedlings.
