墨兰根状茎在气升式反应器中增殖及绿芽分化研究

Rhizome Proliferation of Cymbidium sinense in Air Lift Bioreactor and Shoot Differentiation

摘要: 以墨兰根状茎为外植体,探讨在生物反应器中培养方式、光照强度对墨兰根状茎生长的影响及根状茎外植体长度和部位对绿芽分化的影响。结果表明,气升式生物反应器完全浸没式培养最适合根状茎的增殖生长,光照强度为1600 Lux时根状茎增殖生长最佳。当根状茎外植体长度为1.0 cm时,绿芽分化率、绿芽分化个数和长度都明显高于其他长度的处理;当根状茎外植体部位为底部时,绿芽分化率达到了100 %,绿芽个数和长度也优于其他部位;对绿芽分化过程进行观察,根状茎在接种后第10 d时,表面开始有绿色芽点突起,第20 d之后芽点不再增多但开始不断分化。

Abstract: Rhizomes in vitro of Cymbidium. sinense were used as explants to investigate the effect of culture methods and light intensity on rhizomes of C. sinense in bioreactor, and effects of explant length and part on shoot differentiation. The results showed that immersion culture and light intensity of 1600 Lux of air-lift bioreactor was best for rhizome proliferation and growth. Shoot proliferation rate, shoot number and length were higher in the treatment of 1.0 cm length than others. Bottom part of rhizome explants was more favorable than other parts. The formation progress of green shoots was observed, shoot primordium formed on the surface of rhizome on the 10th, then the primordium never enlarged and development from the 20th.
