牛体细胞核移植技术体系的优化

Optimization of Bovine Somatic Cell Nuclear Transfer Technology System

摘要: 为了提升卵母细胞成熟质量和核移植胚胎的体外发育潜能,在卵母细胞成熟液和胚胎培养液中分别添加不同浓度IGF及胎牛血清FBS,并探讨了二者对牛卵母细胞成熟培养、核移植胚胎发育的影响以及相关性能的变化。结果表明:分别在成熟液中添加40 ng/mL IGF、培养液中添加20% FBS后,成熟率、卵裂率、囊胚率均有显著提高。本研究为今后牛体细胞核移植技术体系的优化和完善打下了坚实的理论基础。

Abstract: To improve the quality of oocyte maturation and developmental competence of nuclear transfer embryos, different concentrations of IGF and FBS were added into oocyte mature medium and embryo nutrient medium, respectively. Then maturation rate, cleavage rate, blastocyst rate were tested to explore the effect of IGF and FBS on bovine oocyte maturation culture and nuclear transfer embryo development. The results showed that adding 40 ng/mL IGF in mature medium and 20% FBS in nutrient medium, maturation rate, cleavage rate, blastocyst rate were significantly increased. The results would lay a foundation for optimizing bovine somatic cell nuclear transfer technology in the future.
