小麦条锈菌DNA提取方法的比较研究

摘要: 以小麦条锈菌夏孢子为实验材料,分析比较了CTAB法、SDS法、尿素法和CTAB/SDS法4种方法对小麦条锈菌基因组DNA的提取效果。结果表明,利用4种方法提取的小麦条锈菌夏孢子DNA的纯度和得率存在一定差异,单位重量夏孢子分离的DNA量依次为:CTAB法>尿素法>CTAB/SDS法>SDS法,而所获得的DNA纯度依次为:CTAB/SDS法>CTAB法>SDS法>尿素法。以不同分离方法获得的小麦条锈菌DNA为模版,利用特异性微卫星引物RJ17进行PCR扩增和电泳分析,4种方法均能满足SSR反应,但CTAB法更适于SSR反应。

Abstract: Uredospores of wheat yellow rust was used as test material. Isolation methods of wheat yellow rust genomic DNA by CTAB,SDS,Urea and CTAB/SDS were compared.The results showed that the purity and yield of wheat yellow rust DNA obtained by above methods were different.These four methods were ranked as: CTAB>Urea>CTAB/SDS>SDS according to the average DNA microgram of uredospores, and ranked as:CTAB/SDS>CTAB>SDS>Urea according to the genomic DNA purity.The DNA obtained by these four methods was subjected to SSR analyses with RJ17 primer ,and the DNA obtained with CTAB method used as template was better than others in SSR analysis.
