南瓜侵染白粉菌后的活性氧爆发观察

Active Oxygen Outbreaks of Pumpkin Infected with Powdery Mildew

摘要:

为探究不同抗性南瓜材料在感染白粉病后活性氧积累水平的变化情况,对中国南瓜自交系112-2和‘九江轿顶’侵染白粉菌后的活性氧爆发情况进行观察。结果表明:在幼苗期,自交系112-2和‘九江轿顶’0 h接种白粉菌孢子,72 h检测到H2O2开始积累,96~144 h H2O2积累量逐步增加。自交系112-2和‘九江轿顶’0 h接种白粉菌孢子,24 h开始检测到O2-,48~144 h O2-积累量逐步增加。在结果期,2种材料在0 h接种白粉菌孢子,48 h自交系112-2开始检测出H2O2,72~144 h H2O2积累量逐渐增多;而‘九江轿顶’72 h开始检测出H2O2,96~144 h H2O2积累量逐渐增多。自交系112-2和‘九江轿顶’0 h接种白粉菌孢子,48 h开始检测到O2-,72~144 h O2-积累量逐步增加。中国南瓜自交系112-2比‘九江轿顶’产生H2O2和O2-的过程迅速,且积累量差异在第144 h达到最大。抗性材料接种白粉菌后H2O2和O2-的积累发生较快,较多于感病材料。

Abstract:

The active oxygen outbreaks of pumpkin inbred line 112-2 and ‘Jiujiangjiaoding’ infected with powdery mildew were observed to explore the correlation of ROS with the resistance of pumpkin to powdery mildew. DAB-dyed spots represented H2O2 accumulation and NBT-dyed spots represented O2- accumulation. The results showed that at the seedling stage, the inbred line 112-2 and ‘Jiujiangjiaoding’ were inoculated with powdery spores at 0 h, and a small amount of DAB-dyed spots were observed at 72 h, and the DAB-dyed spots expanded and deepened from 96 h to 144 h. The inbred line 112-2 and ‘Jiujiangjiaoding’ were inoculated with powdery spores at 0 h, NBT-dyed spots began to appear at 24 h, the NBT-dyed spots continued to expand and deepened from 48 h to 144 h. In the fruiting period, the two materials were inoculated with powdery mildew spores at 0 h, DAB-dyed spots were observed in inbred line 112-2 at 48 h, and the spots enlarged and deepened from 72 h to 144 h; DAB-dyed spots were observed at 72 h from the ‘Jiujiangjiaoding’, and the spots expanded and deepened from 96 h to 144 h. The inbred line 112-2 and ‘Jiujiangjiaoding’ were inoculated with powdery spores at 0 h; NBT-dyed spots began to appear at 48 h, the NBT-dyed spots expand and deepened from 72 h to 144 h. The inbred line 112-2 produced H2O2 and O2- faster than the ‘Jiujiangjiaoding’ and the difference in accumulation amount reached the maximum at 144 h. In conclusion, the accumulation of H2O2 and O2- in the resistant material inoculated with powdery mildew is faster and more than the susceptible material.
