不同品种马铃薯试管薯诱导体系的优化

摘要: 利用5种培养基和3种培养方法对马铃薯的大西洋、陇薯3号和甘农2号品种进行了试管薯的诱导。结果表明:8%的蔗糖浓度是大西洋和陇薯3号试管薯诱导的最佳糖浓度;5mg/L 6-BA的添加可有效提高结薯率和增加块茎直径;在含8%蔗糖的培养基上,琼脂的添加与否对试管薯的诱导率没有影响;在不含蔗糖的MS培养基中,300μmol/L 山梨醇+20mmol/L NH4NO3+20mmol/L KNO3的添加对试管薯的形成和发育不利;3种培养方法中,液体培养法的试管薯诱导效率最好。

Abstract: Potato microtubers of Atlantic, Longshu3 and Gannongshu2 were induced by using 5 mediums and 3 culture methods. The result showed that 8 percent sucrose was the best concentration of inducing Atlantic and Longshu3’s microtuber; Adding 5mg/L 6-BA could improve the rate of microtuber setting and increase microtuber’s diameter; In the 8 percent sucrose medium, agar didn’t affect the rate of microtuber inducing; In the MS medium of no sucrose, 300μmol/L sorbitol added with 20mmol/LNH4NO3 and 20mmol/LKNO3 was harmful to forming and growth of microtuber; In the 3 methods, liquid method was the best method in inducing microtuber.
