MEF2a基因在肌肉组织中的表达研究

Research on Expression of MEF2 Gene in Muscle Tissue

摘要: 分别取90日龄、180日龄和270日龄大白猪心肌、背肌和腿肌,提取总RNA,设计并合成猪MEF2a引物,以β肌动蛋白作为内参,利用Real-time PCR方法检测不同日龄大白猪肌肉组织中MEF2a基因mRNA的表达。结果表明,不同日龄大白猪的心肌和背肌中,MEF2a基因mRNA的表达水平无显著差异,在腿肌中有显著差异(P<0.05)。从90日龄到270日龄,MEF2a基因在心肌、背肌和腿肌的表达水平均差异显著(P<0.05),腿肌最高,其次是背肌,心肌最低。据此推测,MEF2a基因在猪成体后的肌肉组织中,主要是促进Ⅰ型肌纤维的生成,而不再是介导细胞分化。

Abstract: The expression levels of 90d, 180d and 270d Landrace cardiac muscle, dorsal muscle and leg muscle were determined by Real-time PCR.Total RNA was extracted from muscle. The MEF2a PCR primers was designed according to blast homologat the conserved regions.β-actin was used as loading controls.The results showed that the cardiac muscle and dorsal muscle mRNA levels were not significantly varied among different day, expect leg muscle (P<0.05). From 90d to 270d, MEF2a expressions level were significant varied among cardiac muscle, dorsal muscle and leg muscle (P<0.05), and the highest is leg muscle, the second is dorsal muscle, the lowest is cardiac muscle. It was deduced that the main function of MEF2a is to promote I myofibrillar formation and not to mediate cell differentiation during adult pig muscle
