烤烟薄细胞层培养一次成苗与快繁研究

One-step shoot Formation and Micropropagation in the thin cell-layer Cultures of Flue-cured Tobacco

摘要:

烤烟(Nicotiana tabacum L)嫩茎3~15层表皮和亚表皮薄细胞层在附加有适当激素配比的MS培养基上可一次培养成苗。薄细胞层一次培养成苗的适宜培养基为:MS+NAA0.1~0.3 mg/l +IBA0.1 mg/l +6-BA1.5~2.5 mg/l+3%蔗糖;BA浓度控制在2.5mg/l以下有利于培育壮苗。适宜的生根培养基为:1/2MS+2%蔗糖+NAA0.1~0.2 mg/l。

Abstract:

The epidermal and sub- epidermal thin cell-layer of flue-cured tobacco (Nicotiana tabacum L) tender stem in additional appropriate hormone combinations of the MS culture medium can take one-step shoot formation. MS+NAA0.1~0.3 mg/l +IBA0.1 mg/l +6-BA1.5~2.5 mg/l+3%Sucrose culture medium is suitable for one-step shoot formation in thin cell layer cultures. BA concentrations below 2.5mg / l is conducive to nurturing strong seedling. 1/2MS+2% Sucrose +NAA0.1~0.2 mg/l culture medium is suitable for root induction.
