病原侵染后杨树叶片中两个关键酶的表达分析

Expression Analysis of Two Key Enzymes in Poplar Leaves after Pathogen Infection

摘要: 抗坏血酸过氧化物酶(ascorbate peroxidase,APX)和S-腺苷甲硫氨酸合成酶(S-adenosyl methionine synthetase,SAMS)在植物胁迫应答中扮演重要角色。为了研究这两个酶与杨树抗病的关系,以杨树高抗无性系“NL895”(P. euramericana CL“NL895”)为实验材料,利用实时定量RT-PCR(Real-time RT-PCR)技术,分析病原侵染不同时间后杨树叶片中这两个酶在mRNA水平上的表达变化,结果显示,这两个酶均差异表达且表达量下调,表明二者与杨树抗病过程是密切相关的。进一步将APX和SAMS在转录组水平上的表达变化与蛋白质组水平上的变化进行比较后发现,二者在两种水平上的表达变化无显著相关性。

Abstract: Ascorbate peroxidase (APX) and S-adenosyl methionine synthetase (SAMS) play important roles in plant stress response. To study the relations between the two enzymes and poplar disease resistance, the poplar high resistant clone “NL895” was used as the experimental materials and then the expression changes of them at mRNA level after different times of pathogen infection in poplar leaves were analyzed utilizing the real-time RT-PCR. The results showed that the two enzymes were both differential and dropping expression. It indicated that they were closely correlative with poplar disease resistance. It was found that the expression changes of APX and SAMS at transcriptome and proteome levels had no significant correlations by further comparing the expression ones.
