玉米链格孢菌叶枯病病原菌的分子鉴定

摘要:

对玉米链格孢菌叶斑病病原菌进行了分离、纯化,并按照柯赫氏法则证明其为玉米链格孢菌叶斑病病原菌。利用PCR技术对病原菌核糖体DNA内转录间隔区(internal transcribed spacer)进行扩增,并克隆、测序,测序结果显示其与GenBank中报道的细链格孢菌(Alternaria tenuis)同源性为99%,结合形态学特征,确定该病原菌为细链格孢菌。

Abstract:

The pathogen of Maize Alternaria leaf spot was isolated and purified, and according to Koch's postulate, Alternaria leaf spot of maize was caused by the isolated fungi. The internal transcribed spacer (ITS) region of pathogen was amplified by polymerase chain reaction (PCR), and the specific fragment was cloned and sequenced. The result showed that the sequence of ITS for the pathogen shared 99% homology with that of Alternaria tenuis in GenBank. The pathogen was identified as Alternaria tenuis based on the morphological characteristics and sequence of ITS.
