广西黄皮、不同柑桔品种黄龙病检测及16S rDNA序列分析

Detection of Citrus Huanglongbing and 16S rDNA Sequence Analysis in Clausena lansium and Different Citrus arieties from Guangxi

摘要:

运用nested-PCR技术对广西黄皮及11个柑桔品种进行柑桔黄龙病(citrus huanglongbing, HLB)检测,结果表明:从黄皮及11个柑桔品种中均可扩增出一条特异性的400 bp条带,说明广西种植的黄皮及11个柑桔品种均可感染HLB。同时对广西HLB病原菌16S rDNA进行扩增测序,同源性分析和聚类分析表明,广西HLB病原菌属于亚洲种,且在亚洲种不同株系中,与中国厦门株系及日本株系遗传距离较近。

Abstract:

nested-PCR was used to detect citrus Huanglongbing (HLB) in Clausena lansium and 11 citrus varieties. A specific 400 bp band was amplified from plant materials, respectively. The result demonstrated that Clausena lansium and 11 citrus varieties from Guangxi could be infected by HLB. Simultaneously, 16S rDNA of HLB pathogen was amplified and sequenced. Homology analysis and cluster analysis indicated that HLB from Guangxi belonged to ‘Candidatus Liberibacter asiaticus’, and was more closed to Xiamen lines from China and two Japanese lines.
