耐低温兼性厌氧淀粉酶产生菌Y89的筛选及酶学特性

Isolation of the Psychrotolerant Facultative Anoxic Strain Y89 Amylase and Enzymatic Properties

摘要:

从石家庄污水处理厂活性污泥中筛选得到一株兼性产淀粉酶菌株Y89,经形态学、生理生化和16S rDNA序列分析,鉴定为类芽胞杆菌属 (Paenibacillus sp.)。该菌株为耐低温菌,它的最适生长温度为16℃,最适氯化钠浓度5%,最佳pH值为6.0,pH 5.0—9.5范围内均能生长, 培养72-124h为生长稳定期,在40℃进行水浴反应,该菌株酶活较高,为386.13U/ml。

Abstract:

A psychrotolerant strain Y89 producing amylas,which is facultative anoxic , was isolated from activated sludge of sewage treatment plant of ShiJiaZhuang. The morphology, the physiological biochemical characteristics and the 16SrDNA analysis of the strain point out that this organism is a member of the genus Paenibacillus sp.. The strain is psychrotolerant, the optimum growth temperature was 16°C,the NaCl optimum for growth is 5%, the fastest growth occurred within pH6.0, the strain can also grow within a range of 5.0—9.5. The stationary phase of the strain occurred after cultured between72h and 124h ,water bath at 40℃, the amylase displayed maximum activity(386.13U/ml).
