Abstract:
Genomic DNA of Schisandra chinensis (Turcz.) Baill. was extracted by means of three different DNA extraction methods, CTAB, SDS and improved CTAB methods. The purity and quality of DNA obtained by these methods were compared, and the quality was evaluated by agarose gel electrophoresis. The results showed that DNA sample obtained by improved CTAB method had the highest purity and quality. Good repeatability of RAPD amplification can be obtained with this DNA template. Fourteen random primers of good repeatability generated 67 bands, with an average of 4.8 fragments per primer, and 86.6% of them were polymorphic. UPGMA analysis showed similar results with the morphological classification. The accessions can be divided into three groups at the coefficient 0.69. The RAPD technology system can be used to identify 10 Schisandra chinensis (Turcz.) Baill. accessions.
