Abstract:

The aim was to obtain the mutant materials of azuki bean for gene location, clone and gene functional analysis. Dry seeds of azuki bean variety ‘Jingnong 6’ were irradiated with 30, 35, 40, 45, 50, 55 Gy dosage fast neutron respectively for creating new germplasm and obtaining mutants of gene mapping, cloning and functional analysis. Germination rates of M1 generation were 31.72%, 29.06%, 30.29%, 36.51%, 16.15% and 15.17% respectively. Their relative variation frequencies were 3.20%, 4.70%, 6.75%, 6.61%, 4.00%, 2.20%. The higher dosage of fast neutrons was, the lower germination rate was, and the higher variation frequence was relative. The dosages from 40 to 45 Gy were optimistic to azuki bean fast neutron mutagenesis. Seventy seven mutants including leaf shape, plant architecture, seed shape and size, seed coat color, leaf color, early maturity and high yield in M3 generation were gained. These mutants were very useful for gene mapping, gene cloning, further functional analysis and breeding.
