Abstract:
In order to obtain the (Myogenin) MyoG gene sequence of grass carp and its expression pattern in the different muscle tissue. PCR primers were designed according to Cyprinus carpio MyoG gene sequence (AB012881). Using RT-PCR method, we amplified MyoG gene sequence. The analysis and acquired sequences. We also analyzed the mRNA expression character of MyoG gene, using semi-quantitative RT-PCR, in grass carp different tissue. The results showed that grass carp MyoG gene cDNA was 789 bp (GenBank accession No. JQ793897), with an ORF of 762 bp. The cDNA encoded 253 amino acids peptides, with typical MyoD gene family structure trait of Basic helix-loop-helix (b-HLH). Compared with Zebrafish, common carp, rainbow trout and atlantic salmon, the homology of amino acid sequences were higher (73%-95%), but lower in some mammals and poultry (53%-57%). The MyoG gene expression was observed in red muscle, white muscle, fat, hepatopancreas, kidney, brain, intestines, heart and gill and the expression level of MyoG gene in white muscle was significantly higher than that of other tissues studied (P<0.05), and in brain, hepatopancreas and gill also have a high level expression. We successfully obtained the MyoG gene sequence of grass carp, and its expression level in white muscle was significantly higher than that of other tissues. We offered a initial value for the role of MyoG on the muscle tissue development of grass carp.
