Fixative Fluid and Hematoxylin: Effects on Staining Animal Reproductive Organs

Abstract:

To provide a basis for histological staining of different animal reproductive organs, the organs including ovary, testicle, uterine horn and epididymis were collected from small tailed han sheep, Tibetan sheep and yak after slaughtering as test materials, and paraffin sections and hematoxylin-eosin (H-E) staining methods were adopted to observe and compare the dyeing effects of reproductive organs with different fixation solution and hematoxylin staining time. Results showed that the fixation effect with 4% paraformaldehyde for 36 h was similar to that with AAF for 48 h, but the dyeing effect after fixing with AAF showed a distinct advantage, meanwhile the dyeing time after fixing with 4% paraformaldehyde was shortened by 1/2. The study found that, by fixing reproductive organs with different fixed fluids and adjusting the dyeing time of hematoxylin, fixation effects with 4% paraformaldehyde and AAF fixed liquid were different respectively. The time of both fixing the tissues with AAF fixed liquid and dyeing on them with hematoxylin were longer than that of the fixing time on tissue with 4% paraformaldehyde, and the dyeing effect with AAF fixed liquid demonstrated a distinct advantage.
