Diversity of Culturable Endophytic Bacteria from Seeds of Chenopodium quinoa in Xizang

Abstract:

To explore the species and diversity of culturable endophytic bacteria of quinoa seeds in Xizang, and to enrich the resources of endophytic bacteria in quinoa seeds, quinoa seeds were collected from Lhasa, Shigatse, and Nyingchi in Xizang. The species and community structure characteristics of their endophytic bacteria were identified using traditional isolation and cultivation methods. A total of 1,675 strains of endophytic bacteria were obtained through isolation and cultivation. Based on 16S rRNA sequence analysis and morphological characteristics, they were classified into 118 species, 35 genera, 16 families, 7 orders, 4 classes, and 3 phyla. The dominant species were Pantoea agglomerans, Bacillus altitudinis, and Bacillus siamensis. Six types of culture media were used in the experiment, with NA medium showing the highest isolation frequency of 23.1% (387 strains). α-diversity analysis revealed that the culturable endophytic bacteria of quinoa seeds in Lhasa exhibited higher species dominance and evenness, while Nyingchi showed better species richness and relative abundance compared to other two regions. The culturable endophytic bacteria in Xizang quinoa seeds are rich and diverse, demonstrating potential research value.
