Induction of the Microtuber of Dioscorea alata L. in Vitro

Abstract:

By studying the effect of IBA, BA, sucrose, short-illumination and difference temperature of day and night on the induction of microtuber of Dioscorea alata L., to determine the impact factors of the microtuber’s farmation and screening the best training schemes. With Dioscorea alata L. tissue culture in vitro shoot as materials, research the influence of plant hormones, sucrose concentration, the time of light and between day and night processing on Dioscorea alata L. microtuber’s formation. Add plant hormones can promote the Dioscorea alata microtubers formed early, IBA and BA play a role of synergy. It was proved that appropriate sucrose concentration had significant effect on the farmation of Dioscorea alata L. microtubers, no or low level of sucrose causes none forming microtubers and high levels of sucrose lead to inhibition on the growth of Dioscorea alata L.. The suitable concentration that induced more and bigger microtubers was IBA 2.0 mg/L + BA 0.5 mg/L + sucrose 60 g/L. Short light and day and night temperature could also improve the microtuber induction rate, numbers and sizes. Plant hormones, day and light temperature, sucrose concentration were important factors for microtubers formation. The interaction was existed between IBA and BA, and it was affected by sucrose concentration, illumination and temperature conditions. Sucrose existion decided microtubers formation directly.
