Genetic Structure of Patinopecten yessoensis with different shell colours and Correlation of Microsatellite Markers with Growth Traits

Abstract:

Abstract: The ordinary Patinopecten yessoensis (control groups) and "Ivory" strains of Patinopecten yessoensis F2 cultured in Dalian Ocean University, Key Laboratory of Mariculture, Ministry of Agriculture since from 2007 were cultured to adults in Zhang Zidao sea (9-month-old, 60 samples respectively) and used in this experiment, the genetic structure of the two groups were analysed using 22 polymorphic microsatellites markers. The value of He, Na, Ne and, PIC showed that the genetic diversity of the two groups were high and there was no significant difference in genetic structure between the two groups. The band A at locus 8 appeared in most individuals of brown scallops, but it disappeared completely with no amplification in "Ivory" scallops; it is suggested that this band A is possibly the negative genetic marker in white scallops. The SPSS 16.0 was used to analyze the effect of these 23 microsatellites on growth traits. Results indicated that locus HLJX 109, HLJX 178 and HLJX 139 had a significant impact (P<0.05 or P<0.01) on body weight; locus 8, HLJX 191 and HLJX 139 had a significant impact (P<0.05 or P<0.01) on shell length and shell height. This search is working on the relationship between Patinopecten yessoensis’s microsatellites markers and with different shell colours and growth traits in order to provide data for further study on Patinopecten yessoensis’s QTL locations and provide theoretical guidance for Patinopecten yessoensis’s cultivation and breeding.
