Comparative Proteomics Analysis of Rice Seedling for Two Cultivated Varieties of Rice

Abstract:

Rice stripe disease is one of the most economically devastating and widespread viral disease of rice (Oryza sativa L.). To illustrate the molecular mechanism of high resistance rice varieties to rice stripe disease resistance, the two-dimensional electrophoresis and mass spectrometry were used to study the characteristics and differences of protein expression profiles between rice cultivars of ‘Zhendao88’ and ‘Xudao3’, which had high resistance to rice stripe disease. The results showed that approximately 1000 protein spots were separated and visualized on coomassie brilliant blue (CBB) stained two-dimensional electrophoresis (2-DE) gels, which were mainly distributed in the isoelectric point of 4 to 7, 10-100 kDa molecular weight range. With the help of ImageMaster 2D Platinum software, 7 down-regulated proteins were found in ‘Xudao3’ and 6 proteins were ultimately identified by MALDI-TOF. These proteins included energy associated proteins, metabolism associated proteins, defense associated proteins, and unknown protein.
