Studies on Changes of Physiology and Biochemistry Indexes During Adventitious Shoot Regeneration from Stem Explants of Phellodendron amurense

Abstract:

The stem segments were used as explants to study the changes of physiology and biochemistry indexes during adventitious shoot regeneration.The stem segments from aseptic seedling of Phellodendron amurense were inoculated on MS medium containing 1.5 mg/L 6-BA, 0.5 mg/L NAA and 20 g/L glucose. After about 20 days of culture adventitious shoots were induced, and occurred to regenerated plantlets at 30 days or so. By using colorimetry, the SOD activity showed downtrend during the formation of adventitious shoots. While the other antioxidant enzyme (CAT and POD) activities, the concent of soluble protein as well as soluble sugar all increased in varying degrees before peak of adventitious shoot formation, and then decreased. The results showed that there was obvious correlation between antioxidant enzyme activities, the concent of soluble protein and soluble sugar and adventitious shoot regeneration.
