Correlation of Microspore Development Period and the Flower Organ Morphology of White Asparagus

Abstract:

The purpose of this paper is to find a identification method of the developmental asparagus microspore and discuss the relevance between the developmental period of asparagus microspore and the morphology of flower organ. So the phenol magenta, carmine acetate and heidenhain iron hematoxylin solutions were selected for use in staining observation to asparagus microspore, then the developmental cytology, the relevance between the microspore development and the bud’s size have been studied. The results showed that the effect of heidenhain iron hematoxylin was the best in the three solutions. The development of asparagus microspore was divided into tetrad, early- or mid-uninucleate, late-uninucleate and binucleate stages, each stage demonstrated distinctive characters. The developmental periods of asparagus microspore were closely related to the morphological characters of buds. The longitudinal diameter and transverse diameter of buds had significant differences in different developmental periods. When the asparagus microspores were at the late-uninucleate, the buds showed 2.30 mm in longitudinal diameter and 1.76 mm in transverse diameter. The developmental period of asparagus microspore could be judged by the longitudinal diameter and transverse diameter of buds, so as to define the standard of bud selection corresponding with the optimal stage of microspore culture.
