Effects of the Fermentation Medium Components on the Cellulase Production by Cellulomonas X4-9

Abstract:

In this paper, the effects of the components in fermentation medium on the endoglucanase, exoglucanase and β-glucosidase producing by Cellulomonas X4-9 were investigated. The rotational quadratic orthogonal experiment design by DPS soft was used to optimize the fermentation medium (Peanut straw powder, Sucrose, Carbamide, (NH4)2SO4, MgSO4 and KH2PO4) of Cellulomonas X4-9. The results showed that peanut straw powder, sucrose and (NH4)2SO4 had a significantly positive effect on the biosynthesis of the above enzymes, but carbamide, MgSO4 and KH2PO4 caused a negative effect. In the case of the optimal fermentation medium (Peanut straw powder 30 g/L, Sucrose 20 g/L, Carbamide 2.5 g/L, (NH4)2SO4 10 g/L) for Cellulomonas X4-9, the final activity of endoglucanase, eXoglucanase and β-glucosidase was 2.705163, 5.723014 and 3.614921 IU/mL, which was 123.66%, 146.32% and 186.40% higher than that obtained under the original medium recipe, respectively. Cellulomonas X4-9 could produce a group of enzymes including endoglucanase, exoglucanase and β-glucosidase, and the condition of fermentation medium was coincident to the higher productivity.
