Optimization of Deproteinization Process of Polysaccharides in Fruits from Acanthopanax Sessiliflorus Seem.

Abstract:

To determine the best process to remove protein from polysaccharides in Fruits of Acanthopanax Sessiliflorus Seem., two methods including enzymolysis and trichloroacetic acid (TCA)-Sevag were optimized by single factor and orthogonal design. The results showed that polysaccharides solution at pH values adjusted to 6.5 was dealt with the mass concentration of 4% papain for 2 h at 40℃, and then the supernatant following by centrifuged for 10 min at 5000 r/min was handed for four times by 1:1 (v/v) TCA-Sevag [chloroform:butanol=5:1(v/v)] reagent for that of equal volume. The deproteinization rate was 95.6% and the loss rate of polysaccharides was 21.5%. The method of enzymolysis with TCA-Sevag had high efficiency in deproteinization.
