Sodium hypochlorite for sterilization Methods of plant culture medium

Abstract:

he preparation process in the medium instead of sodium hypochlorite added to the medium pressure steam sterilization methods were studied. The result were as following : hypochlorite in the medium added after solidification, 25 ℃, 2000lx light intensity, 16 hours illumination of culture conditions, the concentration of 360mg / L, some parts of the culture medium were still polluted. During the media preparation process, the dissolved agar heated, hot in the medium supplemented with 10mg / L sodium hypochlorite, no polluted culture medium was found. With 'God Ma' Chrysanthemum explants in culture medium during the preparation of the final concentration 20mg / L of sodium hypochlorite,all the plants grew normally, furthermore, their growth conditions were better than that of the group of the high-pressure sterilization. When sodium hypochlorite concentrations was above 50 mg / L, the growth was not only more poor than the control, but also did harm to them. The results show that the proper concentration of sodium hypochlorite instead of high-pressure steam sterilization is effective.
