Study on Tissue Culture Technique in Hemerocallis hybridus ‘Stella D’oro’ Using Ovary as Explants

Abstract:

In order to improve the efficiency of propagation of Hemeroeallis hybridus, this study researched the tissue culture system with ovary as explant in primary culture method; use flower bud of ‘Stella D’oro’ as the test material. The result showed that: the first generation cultivated achieved zero pollution and zero perishing by adopting high density sterilizing buds and the technique of peeling ovary in aseptic, and medium supplemented with 6-BA 2 mg/L+2,4-D 1 mg/L+NAA 0.4 mg/L was the best one for ovary explant callus induction and differentiation. Through studying the basic culture medium, hormone ratio and the highly organic substances of the bud multiplications, it is found that: the basic culture medium B5 was better than MS. The hormone supplemented took effect best with 6-BA 2 mg/L+NAA 0.3 mg/L, the propagation coefficient reached 7.2. Add L-proline and hydrolysis of complex protein on propagation coefficient had little effect on, but, they can effectively improve the growth state of plantlet. The medium supplemented with 1/2MS+0.4 mg/L NAA+20 g/L sucrose was the best for rooting of plantlet. The survival rates of plantlets reached 90% in three conditions: river sand, peaty soil, and 3:1 of vermiculite+peaty soil.
