Study on Phosphate-solubilizing Activity and Suitable Conditions of a Strain of Bacillus mucilaginosus

Abstract: In order to study a strain of Bacillus mucilaginosus’s phosphate-solubilizing activity and its suitable culture conditions for phosphate-solubilizing, Bacillus mucilaginosus was cultured for 7 days in the liquid medium added with lowly soluble calcium phosphate. The content of soluble phosphate in the medium was determined by the means of Molybdenum-Antimony Anti-Spectrophotometric method, and the concentration of cultured cell was measured using dilution plate counting. The effects of carbon source, NaCl concentration, pH value and cell concentration on phosphate-solubilizing activity were analyzed. The results showed that, the phosphate-solubilizing activity of Bacillus mucilaginosus reached the peak when the medium was supplied with the glucose (as carbon source) and 0.03% NaCl in a pH value of 6.0. However the cell concentration had no significant effects on phosphate-solubilizing activity .
