Genetic Analysis of Two Wheat Differential Hosts for Resistance to Yellow Rust

Abstract: According to Puccinia striiformis west isolates virulence spectrum, race 2E16 is used for inoculation when the first leaf is well developed and about 7 cm. in length. The ‘sweep- dusting’ method of inoculation is adopted, giving a very uniform deposit of spores, after that left it in inoculation room act as humidity chambers at (9±2)℃ for 24h and then moved to low temperature greenhouse(day time for 15~19℃,night for 10~14℃) which contribute to the race infection. When the susceptible cultivar Mingxian169 are fully expressed, infection types are investigated and the resistance and susceptible phenotypes of seedlings are evaluated, Chi-square tests are employed to analyze the inheritance of differential host. The result shows that using an assessment of the wheat stripe rust reaction of seedling plants in the F1, F2 and BC1 population of a set of crosses by conventional method between wheat differential host Heines Kolben and winter wheat cultivar Mingxian169, it shows that the proportions of resistant plants obtained following inoculation of F2 populations with race 2E16 indicate that two recessive genes complementarily determine the reactions of Heines Kolben at normal temperature, but for the different crosses between wheat differential host Struba Dickkopf and winter wheat cultivar Mingxian169, it shows that the proportions of resistant plants obtained following inoculation of F2 populations with race 2E16 indicate that single dominant genes determine the reactions of Struba Dickkopf.
