Effects on callus induction and differentiation of different light time, temperature and type of carbohydrate source from Ramie.

Abstract: With Ramie variety of Xiangzhu No.2 as research material, this test studied effects on callus induction and differentiation of different light time, temperature and type of carbohydrate source from Ramie. The results of variance analysis indicated that culture media of glucose as carbohydrate source is superior to of sucrose in induction callus; 12h light/12h darkness treatment benefited to callus growth and all-light was superior to all-darkness treatment. Inducing callus with culture media of glucose as carbohydrate source at 30℃、28℃、25℃ and 23℃ conditions, callus production significantly decreased with temperature lowdown; however inducing callus with culture media of sucrose as carbohydrate source at 30℃、28℃、25℃ and 23℃ conditions, callus production increased within temperature range of 25~28℃ and decreased at a range of over 30℃temperature or down to 23℃. Under light and lower temperature condition callus was easier to differentiate.
