Feeding Bacteria: Effect on Phenoloxidase Activity and Its Gene Expression in Hemolymph of Bombyx mori

Abstract: To investigate the effect of bacteria infection on phenoloxidase activity and its gene expression in hemolymph of Bombyx mori, the real-time fluorescent quantitative PCR is applied to detect the expression of PPO1 and PPO2 in different time (6、12、24、48 h) after feeding Escherichia coli and Staphylococcus aureus, meanwhile the phenoloxidase activity is determined. The results showed that the expression of PPO1 and PPO2 increased at 6 h, 12 h and 24 h after oral infection with the two bacteria, the phenoloxidase activity was significantly higher than that of the control. In addition, the gene expression of PPO1 and PPO2 and the phenoloxidase activity reached peaks at 12 h after Escherichia coli infection, while they reached peaks at 24 h after Staphylococcus aureus infection. The phenoloxidase activity and gene expression were different after Gram positive bacteria and Gram negative bacteria infection, indicating that the immune mechanisms of silkworm induced by different bacteria were different.
