Investigation on the stem base rot of sweetpotato and identification of pathogen species

Abstract: Sweetpotato stem base rot has occurred in the sweetpotato production area in the Taizhou city and its surrounding cities in Zhejiang Province since 2012, caused a significant decline in the local sweetpotato planting area. To identify the occurrence of sweetpotato stem base rot, and pathogenic composition, the field investigation carried out in 11 counties in Zhejiang province from 2015 to 2018, 127 samples were collected for isolation and identification of pathogens. The results showed that there were four main symptoms in the basal rot of sweetpotato in Zhejiang Province. Based on pathogenicity test, morphological characteristics, and sequence analysis of internal transcribed spacer (ITS) region and partial 16S rDNA, 8 pathogens including Dickeya dadantii, Sclerotium rolfsii, Fusarium verticillioides, F. solani, F. oxysporum, Diaporthe batatas, Phomopsis destruens and F. javanicum were identified. The distribution and composition of pathogen in different regions is different, compound infection of multiple pathogens were found to be more common in collected samples of root rot symptom.
