Study on Callus Inducement and Green Shoot Differentiation of White Asparagus Anther

Abstract: Experiments were conducted on the anther of Asparagus officinalis L. to study the callus inducement and green shoot differentiation. Results showed that callus of anther could be induced from the Asparagus anther which had been refrigerated at 4℃ for 4 days and then inoculated to 1/2 MS culture medium of NAA 2.0 mg/L, 6-BA 1.0 mg/L and 6% sucrose. The optimal time to gather anther is April,in which the callus inducement rate arrived at the peak, while in August and November, the rates were lower. Among different breeds and plants, the callus inducement rate of anther varied, in which the breed of ‘UC155’ performed the highest inducement rate (18.9%), distinctively higher than the other three breeds did, including ‘UC142’(11.6%), ‘Gijnlim’(10.3%) and ‘Thielim’(5.8%); Of all the experiment strains, the breed of ‘UC155’-8 showed the highest inducement rate, which was 83.5%. Compared with dark culture, a supplement light of 1000 lx everyday could stimulate the callus inducement of White Asparagus anther. Centrifugal disposal (4000 r/min, 30 min) before inoculation of anther could not increase the inducement rate of callus. The callus which transferred to MS culture medium of NAA 0.5 mg/L, 6-BA 1.0 mg/L and 3% sucrose could differentiate green shoots without root. Differentiation rate varied among different breeds, in which the breed of ‘Gijnlim’ differentiated most (67.2%) and breed ‘UC142’ did least. Part of the non-differentiated callus could differentiate green shoots while transferred to MS culture medium without plant growth regulators.
