Sequence-related Amplified Polymorphism(SRAP):A Novel Technique for Molecular Marker

Abstract: SRAP is a new technique for molecular marker .It is simplicity, reliability, moderate throughput ratio and facile sequencing of selected bands. This paper describes the SRAP principle, protocol. SRAP aimed for amplification of open reading frames(ORFs) by special primer set. The forward prime of 17 base amplified exons, the reverse primer of 18 base amplified introns and promoters. Since introns, promoters and spacers are variable among different individuals, this makes it feasible to generate polymorphism. The amplified DNA fragments are separated by denaturing acrylamide gels and detected by autoradiography. This technique has been used in genetics mapping, genetic diversity, gene tagging, comparative genomics and heterosis prediction. The prospect of the SRAP marker application in plant genetics and research were discussed.
