Abstract: The report on SRAP(Sequence-related amplified polymorphism) analysis of new middle-late maturing lines of rice was rare in China, so the study was done to assess genetic diversity of 16 new lines of Japonica rice in Henan Province based on SRAP. 104 bands were amplified by using 10 pairs of random primers selected from 70 primers, of which 66 were polymorphic bands, and the polymorphic percentage of bands was 63.46%. 8 varieties can be identified by 9 primers which consists of single-marked band or specific-lost band. In addition, 16 accessions were also clustered by SPSS11.5 software. The results showed that 16 accessions could be classified into five groups. Xinfeng 039 and Yujing 6 were clustered respectively. The genetic similarity coefficient of 16 varieties was from 0.186 to 0.808, among which the most insignificant one was Xinjing 03260 and Xindao 03G43, and the most significant one was 3You 121 and Yujing 6. The results at molecular level provided a feasible research foundation for germ plasma identification, genetic diversity analysis and hybrid parents selection in rice breeding.
