The effect of various hormones to induce multiple shoot clumps from axillary bud of Phellodendron amurense and germplasm peservation in vitro

Abstract:

The tender stems of Phellodendron amurense Rupr. was used as explant, studied the effect of various hormones to induce multiple shoot clumps from axillary bud of Phellodendron amurense, root of shoots and germplasm peservation in vitro. Uniform design experiments for screened the suitable medium compositions of in vitro cultured and germplasm peservation in vitro. The results showed that N6+TDZ2.86 mg?L-1 +IAA 0.03 mg?L-1 for axillary bud multiple shoot clumps from tender stem, the rate of induction was 99.5%; 1/2N6+KT 0.45 mg?L-1+IBA 0.10 mg?L-1 for rooting, the rate of rooting was more than 97%; N6+phlorizin2. 65 mg?L-1+KT 0.15 mg?L-1 for germplasm preservation in vitro for 45 months by the retarding of growing at normal temperature, the rate of growth was 0.74%. In vitro culture and germplasm conservation in vitro system of Phellodendron amurense have been successfully established by the methods of inducing axillary bud multiple shoot clumps and the retarding of growing.
