Identification of Lacquer anthracnose pathogen and its biological characteristics research

Abstract:

Lacquer leaves infected with anthracnose disease were collected, and pure culture of the pathogen was acquired by tissue isolation method. The pathogen is determined to be Colletotrichum gloeosporioides (Penz.) Sacc. by its morphologic characters, cultural characteristics, pathogenicity and host range. Further research work on its biological characteristics was conducted. It was found that mycelial growth of Lacquer anthracnose pathogen had a linear correlation with the cultural time. The temperature range of the mycelial growth is 10~35 ℃, and the optimum temperature is 25~28 ℃. The lethal temperature of conidia is 48℃. The conidia cannot germinate until relative humidity reaches 90%. Continuous light may promote the increasing of yield and sporulation of mycelia. The mycelia can grow at the range of pH 3~12, and the optimum growth pH is 6.0. The Lacquer anthracnose pathogen can utilize many kinds of carbon and nitrogen source. The mycelia will grow better if glucose, fruitose was used as the carbon sources and asparamide, alanine was used as the nitrogen sources.
