The Initial Study on Chitinase Producing Strain Fermentation Conditions

Abstract:

In this paper, a chitinase producing strains - Bacillus licheniformis JDZ-3 production of chitinase fermentation conditions were studied. The main factors which determined chitinase production as follows: the optimum initial medium pH was 7.0, the best carbon source in the medium was the colloidal chitin, the best nitrogen source was yeast extract, the best metal ion as Mg2+, the optimal fermentation temperature is 30 ℃. In the orthogonal experiments based on the single factor optimization method, three major nutrients maked up the chitinase fermentation medium were obtained: 0.2% colloidal chitin, yeast extract 0.6%, MgSO4 ? H2O 0.1 %. The strain cultured at 30℃ for 72h with this fermentation medium, the chitinase in the supernatant was up to 2.76U/mL.
