Molecular Identification of Peach Germplasm by ISSR Markers

Abstract:

To differentiate and identify varieties and types of peach germplasm, the genetic diversity and cultivar identification among 28 peach accessions by ISSR markers were explored. Twenty three ISSR primers amplified 188 bands, 96 bands were polymorphic, and the polymorphic percentage was 51.06%. The genetic similarity coefficient (GS) estimated from these molecular data ranged from 0.84 to 0.97, with an average of 0.92. Each genotype had a unique banding profile, and a total of 20 specific ISSR fragments were available to be converted into sequence characterized amplified region (SCAR) or simple sequence repeat (SSR) markers. UPGMA analysis showed that there were not distinct clusters and sub-clusters were consistent with geographical origins and agricultural character of peaches. The results of cluster analysis might be related to the germplasm exchanges between different regions and the characteristics of ISSR markers.
