Fermentation Conditions of a Collagenase-Producing Marine Microorganism

Abstract:

Aeromonas sp.F3 screened from marine mud should secrete a protease to hydrolyze collagen. The fermentation of Aeromonas sp.F3 was investigated through single-factor test. The main factors included fermentation conditions and the medium. With single factor trial, the optimal culture condition was determined: 40℃ of fermentation temperature, pH8 and volume 80~100 mL/250 mL for 24 h. The optimal liquid medium was: 3 g/L glucose as carbon source, 9 g/L yeast extract powder as nitrogen source, 5 g/L NaCl as inorganic salt and 10 mg/L MnSO4 as metal ion. Under the optimum condition, the enzyme activity increased very observably.
