Isolation of Cellulose Bacteria Producing Hydrogen and the Study of Straw Pretreatments

Abstract:

In order to strengthen the effect of degradating straw and turn straw to hydrogen, the author used a modified Hungate anaerobic roll tube method and the disintegration of filter paper method, found a strain which could have better hydrogen production by degradating cellulose, and used 6 kinds of method pretreatments, such as hydrothermal method, the vapor on straw stem exploding and so on to treat straw. The author found a bacterium JYB-13 from already getting strains JYB, JYB-13 could produce hydrogen using amount of carbon sources. Its optimum conditions for hydrogen production temperature of 35℃, pH value of 8.5. Using 6 kinds of method pretreatments, by the effect of producing hydrogen, it was concluded that the strain in 2% sulfuric acid treated straw degradation was the best. After fermentation, the degradation rate of straw was 53.1%, the hydrogen content reached 37.2%. Around the analysis of the composition of straw obtained before and after fermentation, JYB-13 could good use of cellulose and hemicellulose better, and few used lignin.
