不同外植体及植物激素对油茶愈伤组织诱导的影响

Effect of Different Explants and Phytohormone on Callus Induction of Camellia oleifera

摘要:

为了给油茶快速繁殖和生物技术育种提供技术参考,以油茶无菌苗的叶片、茎尖、茎段为材料,研究了外植体及植物激素对愈伤组织的影响。2,4-D与KT的组合更适合于愈伤组织的诱导,叶片、茎段诱导愈伤组织的最佳组合为MS+2,4-D 0.5 mg/L+KT 1.0mg/L,茎尖诱导愈伤组织最佳培养基:MS+2,4-D 0.5 mg/L+KT 2.0 mg/L;在选择的3种外植体中,叶片最容易诱导愈伤组织,诱导率可达到100%。从不同外植体的最高诱导率及对植物激素的需用量来看,3种外植体诱导的难易程度从高到低为:叶片>茎段>茎尖。所以在进行愈伤组织诱导时,应优先选择叶片作为外植体。

Abstract:

The study aimed to provide the technical reference to rapid propagation and biotechnology breeding of Camellia oleifera. Effect of different explants and phytohormone on callus induction of Camellia oleifera was studied by using the asepsis seeding of leaf、stem segments and shoot tip as the explants. Combination of 2,4-D and KT were the best phytohormone for the inducting of callus. In the condition of MS basic medium added with 0.5 mg/L 2,4-D and 1.0 mg/L KT, the callus induction rate were higher when used leaf and stem segments as the explants, while In the condition of MS basic medium added with 0.5 mg/L 2,4-D and 2.0 mg/L KT, the callus induction rate were higher when used leaf and stem segments as the explants. In the three kinds of explants which were selected, leaf was the easiest for inducting, and induction rate could reach 100%. From the induction rate and the amount of plant hormones, leaf is the best explants of the three kinds of explants while inducting callus.
