油茶CoUXS1基因全长cDNA克隆及生物信息学分析

Full-length cDNA Cloning and Bioinformatic Analysis of CoUXS1 Gene in Camellia oleifera

摘要:

为了揭示油茶UDP-葡萄糖醛酸脱羧酶基因的序列特点和结构功能,为该基因的深入研究和开发应用奠定基础。根据油茶种子EST文库中调取的UDP-葡萄糖醛酸脱羧酶基因序列,设计特异引物,利用RT-PCR技术获得该基因全长cDNA克隆,并对其序列进行生物信息学分析。结果表明:该基因阅读框为1032 bp,编码344个氨基酸,具有典型的UXS家族模体序列。与海岛棉GhUXS3基因亲缘关系最近,定位于细胞质,有较强的亲水性,将该基因命名为CoUXS1,GenBank登录号为JN017094。

Abstract:

In order to reveal sequence features and structural function of the UDP-glucuronate decarboxylase gene in Camellia oleifera, and based for the further research, the specific primers were designed according to the UDP-glucuronate decarboxylase gene sequence from Camellia oleifera seed EST library. Using RT-PCR technology, the gene full-length cDNA clones were obtained. Then the sequence was analyzed by bioinformatic software. The results showed that: the ORF box included 1032 bp and encoded 344 amino acids. The sequence had typical UXS family motif sequence. The gene had closest phylogenetic relationship with GhUXS3 gene of Island cotton and localized in the cytoplasm with strong hydrophilicity. The gene was named CoUXS1 and the GenBank accession number was JN017094.
