一种简单快速测定对虾白斑综合症病毒浓度方法的建立

摘要: 测定出对虾白斑综合症病毒(WSSV)中基因组DNA在WSSV总质量的比例,再根据WSSV的基因组大小计算出其质量,计算出完整的WSSV病毒颗粒的质量为3.079×10-15 g。同时测定了各个不同组分在WSSV干重中所占的比例:核衣壳包括基因组DNA和蛋白质,其比例分别为10.17%和43.07%,囊膜包括膜蛋白和各种脂类物质,比例分别为24.76%和22%;测定出WSSV悬液中WSSV浓度与其OD600的换算关系为C=OD600×6.79×108个/μL。

Abstract: First the quality of WSSV genome was calculated and the genomic DNA ratio of total WSSV particle mass was measured. According to the calculation, the mass of complete WSSV particles was 3.079×10-15g. In addition, the different components’proportion in WSSV dry weight was determined. The genomic DNA accounted for 10.17%, nucleocapsid accounted for 43.07%, envelop lipid accounted for 22%, envelop protein accounted for 24.76%. The relation between the concentration of WSSV and the OD600 of the suspension of WSSV is C=OD600×6.79×108virions/μL。
