家蚕脂肪酶(Bmlipase-1)基因在抗血液型脓病不同遗传类型材料中的表达分析及其在育种中的应用

Bmlipase-1 Gene: Expression in Different Genetic Materials Against Nucleopolyhedrovirus and Application in Breeding

摘要: 为了研究家蚕脂肪酶Bmlipase-1 基因表达量与家蚕抗核型多角体病毒病(又称血液型脓病)能力的关联性,以抗性品种‘KNC’和敏感品种‘HYB’及其相互交杂组配材料为试验材料,利用添毒实验与荧光定量PCR技术,检测家蚕不同遗传类型材料在不同感染条件下对血液型脓病的抵抗能力及其相应的Bmlipase-1 基因表达水平。结果表明:Bmlipase-1 表达水平高低与材料的抗性强弱相一致,抗性强的材料的Bmlipase-1 表达量高,反之则低;抗性品种‘KNC’、‘KNC’בHYB’和‘HYB’בKNC’在添毒后的Bmlipase-1 的表达量与未添毒相比有显著上调,而‘HYB’仅微量上调。Bmlipase-1 的表达可以在家蚕育种过程中作为家蚕抗核型多角体病毒材料选择与鉴定的依据之一。

Abstract: In order to study the relation-ship between expression level of Bmlipase-1 and resistance level of silkworm to BmNPV,we took the resistance varieties KNC and sensitive varieties HYB and the hybrid strains of two varieties as materials.Detected the resistance to BmNPV and its corresponding lipase (Bmlipase-1) gene expression level by virus( BmNPV) attack test and fluorescent quantitative PCR(qRT-PCR)technology.The results showed that is consistent with the level of expression of Bmlipase-1 and the strength of resistance. The expression level was higher in silkworm varieties with stronger viral resistance, otherwise it is lower. The expression level of Bmlipase-1 had significant differences between KNC and HYB; After infection by BmNPV ,the expression of Bmlipase-1 in silkworm strains KNC, KNC × HYB and HYB ×KNC was significantly higher than that of non-infection by BmNPV, However, HYB only had trace amounts of regulated changes. It’s as one of bases in select anti-BmNPV material in the process of the silkworm breeding.
