紫外分光光度法测定玉米过氧化氢酶活性新进展

Ultraviolet Spectrophotometry Measurement of Catalase Activity in Maize

摘要: 为简便快速测定植物过氧化氢酶活性,探究过氧化氢酶活性随时间变化的关系,运用紫外分光光度法比较了4 种反应体系中过氧化氢酶(CAT)催化H2O2分解的时间曲线。研究结果表明:CAT催化H2O2的分解反应仅在一定底物浓度范围内符合一级反应,因此计算出速率常数K,即代表CAT活性。直接以0.03% H2O2作为CAT反应液,配制简便,试剂廉价,而且得到的时间曲线效果最好,是CAT反应液的最佳选择。

Abstract: In order to simply and rapidly measure catalase (CAT) activity in plant and explore the correlation between CAT activity and time variation, ultraviolet spectrophotometry was applied to compare the time course of the decomposition of H2O2 catalyzed by CAT in four different reaction systems. The results showed that the decomposed reaction of H2O2 catalyzed by CAT accorded with the first order reaction in a certain substrate concentration. So the constant value of rate K could be represented the activity of CAT. Compared with other reaction solutions, 0.03% H2O2 had such advantages as simple preparation, low cost and could get the best time course in the measurement, so it was the optimum reaction solution of CAT activity measurement.
