摘要: 为了比较福建三明地区不同水稻-烟草轮作土壤细菌多样性及群落结构,认识三明水稻-烟草轮作土壤微生物资源多样性,对不同土壤样品中细菌类群进行分析。通过构建细菌16S rDNA 克隆文库,对将乐砂壤土、将乐壤土、宁化砂壤土和宁化壤土细菌多样性和群落结构进行分析和比较。结果表明:4个土壤样品中的细菌类群包括厚壁菌门(Firmicutes)、变形菌门(Proteobacteria)等11个类群,将乐2个土壤样品中厚壁菌门(Firmicutes)和变形菌门(Proteobacteria)是优势类群;宁化2个土壤样品中变形菌门(Proteobacteria)和酸杆菌门(Acidobacteria)为主要优势菌门。香农-维纳指数(H′)和辛普森指数(D)表明,壤土细菌多样性高于砂壤土。通过构建细菌16S rDNA 克隆文库揭示了三明水稻-烟草轮作土壤中细菌多样性十分丰富,这为初步掌握三明水稻-烟草轮作土壤中细菌种类和组成状况提供了一定的参考依据,也为进一步开发利用该地区水稻-烟草轮作土壤微生物资源提供科学的依据。

Abstract: To compare bacterial diversity and community structure of paddy-tobacco rotation soils in Sanming, Fujian, and to understand microbial diversity resources, bacterial phyla of different soil samples were analyzed. Bacterial 16S rDNA gene libraries of Jiangle sandy loam soil, Jiangle loam soil, Ninghua sandy loam soil and Ninghua loam soil were constructed to study soil bacterial diversity and community structure. The results showed that 11 phyla were detected in bacterial libraries, including Firmicutes and Proteobacteria. Firmicutes and Proteobacteria were the dominant species in the two samples of Jiangle soils, and the dominant species of Ninghua were Proteobacteria and Acidobacteria. The Shannon-Wiener index (H′ ) and Simpson′s diversity index (D ) showed that bacterial diversity of loam soil was higher than that of sandy loam soil. The results of 16S rDNA gene libraries indicated that high bacterial diversity was found in paddy-tobacco rotation soil, the results presented certain references for the preliminary understanding of the bacterial species and composition. At the same time, the study also provided a scientific evidence for making better use of the microbial resources in this area.
